Cell wall residues in yeast protoplast preparations.

چکیده
Protoplast preparations made from Saccharomyces cerevisiae by prolonged treatment with snail digestive juice contained fibrils and chitinous bud-scar residues from the original cell wall.

Protoplast formation and yeast cell-wall structure. The action of the enzymes of the snail, Helix pomatia.
1. The digestive juice of the snail Helix pomatia was used in the study of the degradation of isolated cell-wall preparations from a strain of Saccharomyces carlsbergensis. 2. The crude enzyme system was fractionated by gel filtration and the activities of the more specific fractions thus obtained were examined. 3. Results are discussed with respect to (a) the nature of various factors that are...
متن کاملIsolation and characterization of protoplasts from Saccharomyces rouxii.
Cells of the osmotolerant yeast Saccharomyces rouxii were transformed to protoplasts in good yield (85%) by digesting cell walls with snail-gut enzyme in the presence of 10 mM dithioerythritol, 0.1 M sodium phosphate buffer (pH 6.8), and 2.0 M KCl. The requirement for 2.0 M KCl compares with that for S. bisporus var. mellis (another osmotolerant species) and contrasts with the 0.3 to 0.8 M KCl ...
متن کاملDielectrophoretic effect of nonuniform electric fields on the protoplast cell
In recent years, dielectrophoresis based microfluidics systems have been used to manipulate colloids, inert particles, and biological microparticles, such as red blood cells, white blood cells, platelets, cancer cells, bacteria, yeast, microorganisms, proteins, DNA, etc. In the current study the governing electric potential equations have been solved in the presence of cell for the purpose of ...
متن کاملPorosity of the yeast cell wall and membrane.
The limiting sizes of molecules that can permeate the intact cell wall and protoplast membrane of Saccharomyces cerevisiae were determined from the inflection points in a triphasic pattern of passive equilibrium uptake values obtained with a series of inert probing molecules varying in molecular size. In the phase identified with the yeast protoplast, the uptake-exclusion threshold corresponded...
متن کاملSurface structure of yeast protoplasts.
The fine structure of the yeast cell wall during protoplast formation was studied by means of phase-contrast microscopy and the freeze-etching technique. The freeze-etching results indicated that at least in some cases the entire wall substance was not removed from the surface of the protoplasts. After a treatment of 30 min to 3 hr with 2% snail enzymes, an innermost thin wall layer as well as ...
